بررسی بیان ژن و خالص سازی زیر واحدهای S100 A8 , S100 A9 کالپروتکتین و بررسی ساختارآنها

Abstract:
زمینه و هدف: کالپروتکتین، S100 A8 و S100 A9 در فرآیندهای مهمی نظیر پیام رسانی و تنظیم پاسخ های التهابی نقش دارند. در این مطالعه، بیان S100 A8 و S100 A9 بصورت نوترکیب، خالص سازی و بررسی ساختار آنها انجام گردید. روش کار: در این مطالعه تجربی از pET15b به عنوان حامل توالی کد کننده ژنهای S100 A8, S100 A9 انسانی و از ( E.coli BL21 (DE3 به عنوان میزبان استفاده گردید. بیان ژن و فرآیند خالص سازی از طریق SDS-PAGE مورد ارزیابی قرارگرفت. خالص سازی پروتئینها با استفاده از رزین Ni-NTA و بر اساس میل ترکیبی آن به His-Tag پروتئین های نوترکیب انجام گرفت. ساختار سوم پروتئین ها با طیف سنج فلورسانس بررسی شد. یافته ها: زیر واحدهای مذکور 4-3 ساعت بعد از القاء و دمای 37 درجه سانتی گراد بیان بالایی را نشان دادند. درصد بالایی از پروتئین S100A9 در فاز اینکلوژن بادی نشان داده شد، در حالیکه زیر واحد S100A8 عمدتا بصورت محلول بیان گردید. S100A8 و S100A9 در غلظت mM 100 ایمیدازول خالص سازی گردید. در بررسی طیف سنجی نشان داده شد که اسید آمینه تریپتوفان در ساختار های داخلی قرار گرفته و کمتر در معرض محیط آبی است. نتیجه گیری: در این مطالعه زیر واحدهای S100A8 و S100A9 بصورت نوترکیب بیان شده و تخلیص گردید. همچنین، ساختار آنها مورد تایید قرار گرفت.
